panget na pGlo

BAKIT BA KASI?????????????

HA!

Anong bang pakialam ng replication rate o copy number sa arabinose, ha?!

bakit ba masmadaming plasmid sa lb amp ara?!

o masmarami nga ba?

e bakit pa nagLB amp ara solution? bakit hindi lb amp?

GRABEEEEEEEEEEEEEEEEEEE

anyway, next question na nga muna :D wala kong naaachieve dito haha

Hi Tin! :)

I hope the stress of the lab rep isn't affecting you too much.:) I'm sure if you look for the answers, you'd find them in time.:)

Anyway, I was very very frustrated with the same question too.:) But I made a quick search and I think I found the answer.:) pGLO has reporter genes including an ampicillin resistance gene and a GFP gene.:) But the GFP gene is only expressed when pGLO's ara gene is expressed in the presence of arabinose, making the colonies green.:)

But that doesn't answer your question.:) Colonies with pGLO plasmid can grow in the LB amp and LB amp/ara plates.:) And they'd grow at the same rate.:) It's just that the colonies in the LB amp/ara plate will be green under UV.:) I think it just gives you a super sure way of knowing the colony you have has the plasmid (although growing on the LB amp plate is already enough). So just to be super sure.:)

Here's my source of equal growth rates from Biorad: http://www.bio-rad.com/LifeScience/pdf/Biobusiness.pdf

God bless you Tin!:) Hope it works out.:) Yup, nakakastress talaga ang labrep.:)
